ANALYSES OF GENETIC VARIABILITY IN LENTINULA EDODES THROUGH MYCELIA RESPONSES TO DIFFERENT ABIOTIC CONDITIONS AND RAPD MOLECULAR MARKERS BJM

The growth of thirty-four Lentinula edodes strains submitted to different mycelial cultivation conditions (pH and temperature) was evaluated and strain variability was assessed by RAPD molecular markers. The growth at three pH values (5, 6 and 7) and four different temperatures (16, 25, 28 and 37ºC) was measured using the in vitro mycelial development rate and water retention as parameters. Mycelial cultivation was successful at all pH tested, while the ideal temperature for mycelial cultivation ranged between 25 and 28ºC. The water content was lower in strains grown at 37ºC. ANTAGONISTIC EFFECT OF EDIBLE MUSHROOM EXTRACT ON CANDIDA ALBICANS GROWTH BJM

Five species of edible mushrooms, Lentinula edodes, Pleurotus ostreatus, Pholiota nameko, Macrolepiota bonaerensis and Agaricus blazei, were tested for their potential to inhibit the in vitro growth of the pathogenic yeast Candida albicans. Only L. edodes had a fungistatic effect on this human pathogen. The inhibitory compound was produced intra and extracellularly in submersed L. edodes culture, and was also present in fresh and dehydrated mushroom basidiocarps. The fungistatic compound was heat sensitive and lost activity after 72 hours.

Low carbon/nitrogen ratio increases laccase production from basidiomycetes in solid substrate cultivation Scientia Agricola

Basidiomycetes are laccase producers used for hydrolysis of lignocellulosic byproducts in fermentative processes and could be used on biofuel production or ruminant feeding. The objective of this study was to evaluate the effect of concentrations of non-protein nitrogen sources on laccase production and mycelial growth of Pleurotus ostreatus, Lentinula edodes and Agaricus blazei. The fungi were grown on soybean hulls to which urea (U), ammonium sulfate (AS) or mixture of AS:U (1:1) were added to achieve carbon/nitrogen (C/N) ratios of 5, 15, 20 or 30. Morphological and molecular identification of four Brazilian commercial isolates of Pleurotus spp. and cultivation on corncob BABT

The species of Pleurotus have great commercial importance and adaptability for growth and fructification within a wide variety of agro-industrial lignocellulosic wastes. In this study, two substrates prepared from ground corncobs supplemented with rice bran and charcoal were tested for mycelium growth kinetics in test tubes and for the cultivation of four Pleurotus commercial isolates in polypropylene bags. The identification of the isolates was based on the morphology of the basidiomata obtained and on sequencing of the LSU rDNA gene. Novel insights into the early stages of infection by oval conidia of Colletotrichum sublineolum Scientia Agricola

Anthracnose, caused by Colletotrichum sublineolum Henn. ex Sacc. & Trotter, is one of the most important sorghum [Sorghum bicolor (L.) Moench] diseases in Brazil. This fungus exhibits conidial dimorphism, producing either falcate or oval conidia on solid and liquid media, respectively. We compared patterns of the initial infection events by these two types of conidia on sorghum leaves using light microscopy and scanning electron microscopy. The infection events during the first 24 h were similar for both oval and falcate conidia. Globose and melanized apressoria were formed at 24 h after inoculation (hai) regardless of the conidia type. Obtenção de linhagens de Lentinula edodes resistentes a temperaturas elevadas e seleção de linhagens resistentes ao Trichoderma sp Ciência e Agrotecnologia

Lentinula edodes (Berk.) Pegler (shiitake) é o segundo cogumelo mais consumido no mundo e sua produção pode ser feita em toras de madeira ou em cultivo axênico utilizando resíduos agroindustriais como substrato. O cultivo do shiitake no Brasil tem sido gravemente prejudicado pela alta incidência de fungos contaminantes, como Trichoderma sp., e pela ausência de linhagens mais adaptadas às temperaturas tropicais. Duas linhagens resistentes ao Trichoderma sp. foram obtidas por meio de seleção quando crescidas em contato com o fungo contaminante e, por meio de fragmentação micelial, foram obtidas linhagens resistentes a temperaturas elevadas, capazes de crescer a 30º C em meio de cultura.

The biology and potential for genetic research of transposable elements in filamentous fungi Genet. Mol. Biol.

Recently many transposable elements have been identified and characterized in filamentous fungi, especially in species of agricultural, biotechnological and medical interest. Similar to the elements found in other eukaryotes, fungal transposons can be classified as class I elements (retrotransposons) that use RNA and reverse transcriptase and class II elements (DNA transposons) that use DNA. The changes (transposition and recombination) caused by transposons can supply wide-ranging genetic variation, especially for species that do not have a sexual phase. The application of transposable elements to gene isolation and population analysis is an important tool for molecular biology and studies of fungal evolution.

Use of oval conidia as a tool to assess the genetic transfer among Colletotrichum sublineolum mutants BABT

The aim of this study was to assess oval conidia production in mutants for the reductase nitrate gene in C. sublineolum and verify the possibility of using them as a tool to transfer the genetic material in this species. The mutants used in the present study lost the ability to form falcate conidia but all produced oval conidia. The number of nuclei by conidium was evaluated. Oval conidia were efficient in the heterokaryon formation and these heterokaryons in liquid culture medium they also produced oval conidia in abundance. Recombinants were obtained and the genetic exchanges were confirmed by the RAPD analyses.
